Activation of orexin/hypocretin type 1 receptors stimulates cAMP synthesis in primary cultures of rat astrocytes.
Journal Title: Pharmacological Reports - Year 2011, Vol 63, Issue 3
Abstract
The effects of orexins, which are also named hypocretins, on cAMP formation were examined in primary cultures of rat astrocytes. Orexin A, an agonist of OX(1) and OX(2) receptors, stimulated cAMP production with an EC(50) value of 0.68 μM and potentiated the forskolin-induced increase in the nucleotide synthesis. [Ala(11)-D-Leu(15)]orexin B, an agonist of OX(2) receptors, was inactive. The effects of orexin A were antagonized by SB 408124, a selective blocker of OX(1) receptors, but were not affected by TCS OX2 29, a selective antagonist of OX(3) receptors. We hypothesized that the activation of OX(1) receptors stimulated cAMP synthesis in primary rat astrocyte cultures.

Acute doxorubicin pulmotoxicity in rats with malignant neoplasm is effectively treated with fullerenol C60(OH)24 through inhibition of oxidative stress.
The aim of this study was to investigate the possible protective role of fullerenol (FLR, C(60)(OH)(24) on doxorubicin (DOX)-induced lung toxicity using biochemical and histopathological approaches. Rats (Sprague-Dawley...
Pharmacokinetics and ocular disposition of paracetamol and paracetamol glucuronide in rabbits with diabetes mellitus induced by alloxan.
Background: This study evaluates the pharmacokinetics (PK) and ocular disposition of paracetamol and paracetamol glucuronide in diabetic rabbits. Methods: Thirty two New Zealand rabbits were divided into four groups: con...
Modulatory effects of sesamol in dinitrochlorobenzene-induced inflammatory bowel disorder in albino rats.
Background: Inflammatory bowel disease (IBD) is a chronic inflammatory condition of gastrointestinal tract of immune, genetic and environmental origin. In the present study, we examined the effect of sesamol (SES), the m...
Neonatal N-(-2-chloroethyl)-N-ethyl-2-bromobenzylamine (DSP-4) treatment modifies the vulnerability to phenobarbital- and ethanol-evoked sedative-hypnotic effects in adult rats.
To study the influence of the central noradrenergic system on sensitivity to sedative-hypnotic effects mediated by the aminobutyric acid (GABA) system, intact rats were contrasted with rats in which noradrenergic nerves...
Chemical structure of phenothiazines and their biological activity.
Phenothiazines belong to the oldest, synthetic antipsychotic drugs, which do not have their precursor in the world of natural compounds. Apart from their fundamental neuroleptic action connected with the dopaminergic rec...
