Automated system for counting and sorting cells in sugarcane suspensions

Abstract

The quantity and types (embryogenic and non-embryogenic) of cells are very important parameters to evaluate plant cell suspension growth for determine the best suspension to obtaining plant regeneration by somatic embryogenesis. These parameters are determined normally by visual human inspection in the microscope. It provokes fatigue and time lost to biologist and non-precise results. By solve these problems an automatic system was implemented. A CCD-camera is attached at the microscope and a 256 grey level image is digitised. The cell clusters segmentation and position in the image are calculated using the competitive learning and contour tracking technique. The cell cluster with a little starch content are counting and classify by the topological and directional information extracted of cell cluster skeleton. The cell clusters with high starch content are counting and classify by the critical point of cluster contour. The system was validate in Sugar-cane suspension (Saccharum sp.). var. CP 52-43. The final result obtained by human visual inspection and by the automatic system implemented by us was found similar.
