Changes in Proteins in Aqueous Humor of Glaucoma Patients Compared to Cataract Patients

Abstract
Purpose: The Aqueos Humour (AH) is well accessible and provides nutrition and immunity in the anterior segment of the eye. Purpose of our study was to uncover glaucoma related proteomic changes in AH. Patients and Methods: AH samples from 35 patients (Controls: n=12, age=67.4±13.6; POAG: n=23, age: 72.5±8.3) were obtained during cataract or glaucoma surgery. AH samples were subsequently pooled into the experimental groups under equal contribution in terms of protein amount of each individual patient. Protein samples were analysed by LTQ Orbitrap MS device with an upstream LC system. The obtained raw data were analysed using the Maxquant proteome software. Proteins with a fold-change ratio up-regulation higher than a cut-off of 2 were considered as noticeably altered. Results: Quantitative mass spectrometric analysis of AH from POAG- and cataractpatients identified a total number of 175 proteins. Apolipoprotein D (3.16), complement C3 (2.96) pigment epithelium-derived factor (fold change 2.86), dickkopf-related protein 3 (fold change 2.18), Wnt inhibitory factor 1(fold change 2.35) were significantly changed within the AH of glaucoma patients.Glaucoma is a leading cause for vision loss and blindness worldwide. Glaucoma is characterized by accelerated Retinal Ganglion Cell (RGC) and axonal degeneration, which results in progressing irreversible visual loss and ultimately blindness in patients. Elevated Intraocular Pressure (IOP) is the main risk factor and controlling IOP is by far the only efficient treatment in clinical practice [1]. The stabilization of IOP is majorly depend on the balance of production and drainage of Aqueous Humour (AH). The accumulation of excessive AH amounts leads to increased IOP. Besides this, AH also provides nutrition and immunity in the anterior segment of the eye. Its protein composition alters depending on various ocular conditions. Even though, the exact pathogenesis of glaucoma remains obscure, the change in AH protein may provide an insight into involved molecular mechanisms of glaucoma, and its early diagnosis, prediction of prognosis, and patients’ responses to treatments. Proteomics as a rising technique for biomarker discovery, is used in this study to uncover glaucoma related proteomic changes in AH. Purpose of our study was to uncover glaucoma related proteomic changes in AH.
