Cloning and Sequence Comparison of Phytase (Phy) Genes from Aspergillus niger and Bacillus atrophaeus
Journal Title: Annual Research & Review in Biology - Year 2017, Vol 16, Issue 1
Abstract
Aims: To clone phytase genes from prokaryotic and eukaryotic microorganisms and provide partial sequence comparisons between the two systems in relative to their protein functional domains. Study Design: Gene cloning of Phy genes from Bacillus atrophaeus and Aspergillus niger strain 103 and analysis of their amino acid sequence. Methodology: Genomic DNA was isolated from microbial prokaryotic and eukaryotic Bacillus atrophaeus and Aspergillus niger strain 103, respectively. Their phytase genes were isolated and cloned, sequenced and translated into amino acid sequences. Their genes were analyzed and compared with their homologues that are available on the GenBank database using appropriated bioinformatics tools. Results: This study reports the successful cloning of phytase genes from Bacillus atrophaeus and Aspergillus niger strain 103 and the partial characterization and comparison of their sequences in relative to conserved domains that may affect their enzymatic activities. Both genes possess the required phytase activity domains even though no significant homology exists between the bacterial and fungal Phy genes. Conclusion: Phytase genes possess a wide spectrum of hydrolyzing enzyme structures that support the required ability of the enzyme to degrade phytate.

European Sea Bass (Dicentrarchus labrax L. 1758) As a Sentinel Species in Europe to Study the Effects of Contaminants
Aim: The current study was designed to evaluate whether European Sea Bass (Dicentrarchus labrax L. 1758) can be used as a sentinel species to test contaminants. For this aim the acute lethality (96 h) of anionic surfacta...
Rheological Properties of Erythrocytes of Healthy Piglets during the Transition from Dairy to Vegetable Nutrition
In the course of ontogenesis mammals can experience some changes of regular elements’ micro rheological peculiarities. These changes can influence blood rheological properties in capillaries and, so, the degree of tissue...
Comparative Analysis of Contaminability between Clarias gariepinus and Tilapia mariae
Water, Clarias gariepinus and Tilapia mariae samples were collected from four (4) stations across the length of the Osse River between the periods of April, 2013 to September, 2014. Heavy metals (Iron, manganese, nickel...
Water Stress Alleviation of Roselle Plant by Silicon Treatment Through Some Physiological and Biochemical Responses
A research trial was planned to find out the effect of water deficit and/or silicon (Si) foliar application on the growth, yield, sepal quality and some physiological characters of roselle plants. Under water stress trea...
