Cloning, expression and purification of resistance gene analogue RGPM 301 from pearl millet in Escherichia coli
Journal Title: Journal of Applied Biology & Biotechnology - Year 2016, Vol 4, Issue 2
Abstract
Plants combat their pathogens with an array of defense responses. One of the key mechanisms involves products of resistance (R) genes which are responsible for recognition of effector molecules from pathogens and subsequent triggering of defense responses. Resistance gene analogues (RGAs) containing the specific conserved domains of R-genes are isolated from various plants using degenerate oligonucleotide primer based PCR approach. In an earlier study, RGPM 301 an RGA from pearl millet shown to be involved in resistance mechanism against downy mildew disease was isolated and characterized. In the present study, RGPM 301 containing an open reading frame (ORF) of 992 amino acids was cloned into pRSET A expression vector and expressed in Escherichia coli as a Hig-tag fusion protein. The recombinant RGA RGPM 301 was purified to near homogeneity using the Nickel-CL agarose column. Its molecular mass was found to be 120 kDa when separated on the SDS-PAGE which was confirmed by western blotting analysis using the anti-His antibody. The purified protein was subjected to in-gel trypsin digestion followed by mass spectrometric analysis for the confirmation of its identity. These findings facilitate further studies on the exact role of this RGA in the pearl millet downy mildew host pathogen system.
Control of Fusarium oxysporum infection in transgenic tobacco carrying oxalate descarboxilase gene
Fusarium oxysporum is a widespread necrotrophic plant pathogen. Its infection affects several crop plants such as potato, tomato, cotton, banana, coffee, strawberry and sugarcane. The fungus produces oxalic acid, which t...

Klebsiella species known to exhibit important PGP traits like solubilization of phosphate, phytohormone production and good germination potential. In present study, based on the Phosphate solubilization and IAA productio...
Influence of growth conditions on production of poly(3-hydroxybutyrate) by Bacillus cereus HAL 03 endophytic to Helianthus annuus L.
Systematic screening of culturable endophytic bacteria for production of poly(3-hydroxybutyric acid) [P(3HB)] has resulted in the isolation of a potent strain identified as Bacillus cereus HAL 03 (GenBank Accession No. K...
Morphological and Elemental analysis of the effluent of Lead-acid battery manufacturing
Excessive release of Lead (Pb) contaminated effluent into the water bodies become the lead poisonous. This waste water is the main source for the transfer of Lead (Pb) into the living organisms. Lead (Pb) is a poisonous...
Phenotypic and genotypic diversity of Xanthomonas axonopodis pv. manihotis causing bacterial blight disease of cassava in Kenya
Cassava bacterial blight (CBB), caused by Xanthomonas axonopodis pv. manihotis (Xam), is the most important bacterial disease of cassava. There is no information available on the morphological and genetic variability of...
