Cyto- and genotoxicity of selected components of composite materials and their bonding systems

Abstract

Introduction: Results of many studies indicate that 2-hydroxyethyl methacrylate (HEMA) and triethyleneglycol dimethacrylate (TEGDMA) may interfere with cellular homeostasis by interacting with cellular membrane or depleting intracellular level of glutathione. Both monomers increase the level of reactive oxygen species (ROS), and, in consequence, disturb the intracellular redox balance. This process can result in apoptosis induction or cell cycle delay. Methacrylate monomers can also interact with DNA. HEMA and TEGDMA can induce the formation of micronuclei and alter expression of the genes whose products are involved in the regulation of cell growth, gene expression, proliferation and differentiation, cell death, replication and DNA repair.Aim of the study: To discuss processes responsible for methacrylate monomer releasing from resin-based dental fillings, and to review mechanisms of HEMA and TEGDMA cytotoxicity and genotoxicity.Conclusion: Understanding mechanisms responsible for methacrylate monomer toxicity may help explain how these chemicals affect the human body, while improving their physical and chemical properties may contribute to protecting patients’ health.
