DESIGN AND EVALUATION OF SUSTAINED RELEASE CAPSULE FOR ACETAZOLAMIDE

Abstract
The oral route of drug delivery is typically considered the preferred and most patient-convenient means of drug administration. The advances in the formulation technology of modified release dosage form with sustained release oral dosage form has been widely accepted approach as compared to conventional immediate release formulations of the same drug, over which it provides a prolong release of the drug over extended period of time there by giving the better patient compliance and enhanced bioavailability and resulting blood concentration-time profiles of drugs that otherwise suffer from few limitations. Sustain release system are considered a wiser approach for the drugs with short half-lives and which require repeated dosing, they are easy to formulate and are irrespective of absorption process from gastrointestinal tract after oral administration. Acetazolamide is an inhibitor of carbonic anhydrase and is used mainly in the management of glaucoma. It is also used in the treatment of various forms of epilepsy, to prevent or ameliorate the symptoms of acute high altitude sickness and in the promotion of diuresis in instances of abnormal fluid retention, for example, cardiac edema. Acetazolamide is a drug having a shorter half-life of 4 hours. Hence, in present study we have prepared a sustained release capsule of Acetazolamide by using various polymers like HPMC K100M, Eudragit NE30D, HPMC K4M, MCC PH101, ethyl cellulose, carbopol 971P. Matrix embedding method was adopted to formulate sustained release capsule of acetazolamide. The formulation with 8.01% HPMC K100M, 0.68% talc and 4.18% Eudragit NE 30D coating was consider as a best product with respect to in-vitro drug release study.
