Development of antiasthmatic polyherbal formulation (triratna) as an alternative to synthetic steroidal formulation

Abstract

In this study, methanolic leaves extract of Physalis angulata Linn (Solanaceae) (PAL), Adhatodavasaka Linn (Acanthaceae) (AVL) and Solanum nigrum Linn (Solanaceae) (SNL) and formulation (Triratna) of these three extracts were studied for antihistaminic properties by using isolated animal smooth muscle models to find out an alternative to synthetic steroidal formulation, where PAL possesses inhibitory efficacy by 99.58% (1Bg), 101.66% (2Bg), 102.08% (4Bg) in Guinea Pig ileum preparation (GPIP) against 1μg histamine; 100% (1Bg), 133% (2Bg), 126% (4Bg) in Guinea Pig tracheal chain preparation (GPTCP)against 1μg histamine and 50% (1Bg), 75% (2Bg), 100% (4Bg)in fundus strip preparation (FSP) against 1μg 5HT. AVL possesses inhibitory efficacy by 96.66%(1Bg), 97.5% (2Bg), 101.25%(4Bg) in GPIP against 1μg histamine, 46.66% (1Bg), 60% (2Bg),120% (4Bg) in GPTCP against 1μg histamine and 75% (1Bg), 100% (2Bg), 102.5% (4Bg) in FSP against 1μg 5HT. SNL posses inhibitory efficacy by 98.33% (1Bg), 101.25% (2Bg), 102.08% (4Bg) in GPIP against 1μg histamine, 73.33% (1Bg), 100% (2Bg), 120% (4Bg) in GPTCP against 1μghistamine and 25% (1Bg), 50% (2Bg), 75% (4Bg) in FSP against 1μg 5HT. While formulation possesses synergetic effect viz.125% (1Bg), 141% (2Bg) and 153% (4Bg) of inhibition against (1Bg) histamine in GPIP and 73%(1Bg), 105% (2Bg) and 115% (4Bg) of inhibition against 1 Bg histamine in GPTCP. Acute toxicity study of formulation also performed to measure the safety prospective of the formulation that confirmed safety measure of the formulation. Triratna could be novel anti antiasthmatic formulation for next generation.
