Dynamic functional connectivety study of rapid anti-depression and suicidal ideation relief with SAINT

Abstract
Objective To analyze the dynamic functional connectivity ( DFC) characteristics of brain networks in patients with major depressive disorder ( MDD ) treated by Stanford Accelerated Intelligent Neuromodulation Therapy (SAINT), combined with the sliding window method. Methods This paper combined the dynamic method of DFCN sliding window method and K-means clustering to study the DFC changes of neural circuits of rapid anti-depression and suicidal ideation relief with SAINT, and explored the dynamic network connectivity changes through the correlation analysis of neuroimaging data and clinical characteristics, Beck Scale for Suicide Ideation-Chinese Version and Hamilton Depression Rating Scale (HAMD-17). Results The clustering results showed that state I with positive intensity and low frequency, state Ⅱ with negative intensity and high frequency, the number of state transitions and score windows of DFC indexes were significantly different before and after treatment, mean dwell time state I and stateⅡ were negatively correlated with HAMD-17, and the analysis found that depressive symptoms were closely related to the high frequency sparse networks. Nodes of inferior frontal gyrus ( IFG), insular gyrus ( INS), cingulate gyrus ( CG), basal ganglia ( BG), and lateral occipital cortex (LOcC) showed altered functional connectivity strength and variability. Conclusion Abnormalities in the DFC of IFG, INS, CG, BG, and LOcC circuits in MDD patients after SAINT treatment may provide new enlightenments to explore the underlying mechanisms of SAINT.
