Effectiveness of incipient caries induction in microaerophilic and/or aerobic conditions using Streptococcus mutans and Lactobacillus acidophilus. In vitro studies.

Abstract
Introduction. So far various chemical and microbiological models have been used to induce in vitro caries. Demineralizing agents (e.g. acids) caused only surface demineralization. Enamel demineralization at early stage of in vivo caries process has subsurface character. Thanks to microbiological models, using single or poly-bacterial cultures, various environments and different process durations, the development of caries which resemble in vivo stains is possible. Aim of the study. Comparison of effectiveness in in vitro obtainment of incipient caries, using Streptococcus mutans and Lactobacillus acidophilus incubated in BHI and PP media supplemented with 5% sucrose under microaerophilic and/or aerobic conditions. Materials and methods. Experiments were performed on 50 intact teeth coated with human saliva. 25 teeth were incubated in BHI with sucrose and S. mutans (1 month), then with L. acidophilus (3 months) under aerobic conditions at 37°C (Method I). Remaining 25 teeth were incubated in PP with sucrose in the presence of S. mutans in microaerophilic environment (1 month) and then with L. acidophilus under aerobic conditions (3 months) at 37°C (Method II). The enamel lesions were evaluated using visual, probing, SEM and microtomography inspections after 2, 4, 6, 8, 10, 12, 14 and 16 weeks of incubation, respectively. Results. In both methods caries incipiens were obtained. However, in Method II (PP with sucrose, variable conditions of the incubation - microaerophilic and aerobic) caries lesions occurred earlier, were more distinct and corresponded to those observed in vivo. Conclusion. Microbiological Method II, using S. mutans and L. acidophilus, proved more efficient in caries-like enamel lesions induction.<br/><br/>
