In situ genomic DNA extraction for PCR analysis of regions of interest in four plant species and one filamentous fungi
Journal Title: Biotecnología Vegetal - Year 2014, Vol 14, Issue 3
Abstract
The extraction methods of genomic DNA are usually laborious and hazardous to human health and the environment by the use of organic solvents (chloroform and phenol). In this work a protocol for in situ extraction of genomic DNA by alkaline lysis is validated. It was used in order to amplify regions of DNA in four species of plants and fungi by polymerase chain reaction (PCR). From plant material of Saccharum officinarum L., Carica papaya L. and Digitalis purpurea L. it was possible to extend different regions of the genome through PCR. Furthermore, it was possible to amplify a fragment of avr-4 gene DNA purified from lyophilized mycelium of Mycosphaerella fijiensis. Additionally, it was possible to amplify the region ap24 transgene inserted into the genome of banana cv. ‘Grande naine’ (Musa AAA).

In vitro multiplication of Zantedeschia spp. variety Treasure in Temporary Immersion Systems
Callas (Zantedeschia spp.) are plants from Araceae family of a great interest as ornamental plants in pots and as cut flowers. Although they have been propagated by in vitro culture, there are few scientific studies on...
Effect of foliar fertilization on Ananas comosus L. Merr. cv. ‘Cayena lisa’ acclimatization
The low survival and slow growth of in vitro pineapple plants (Ananas comosus L. Merr.) in acclimatization stage limit the use of biotechnological techniques for it propagation. The aim of this study was to determine t...
Production of cardiotonic glycosides in Digitalis purpurea using temporary immersion systems
Digitalis purpurea, is an example of cardiotonic drug. Digoxin and digitoxin are glycosides produced by this plant which are cardiac stimulants with a wide use in the treatment of heart problems. The objective of this...
Characterization of sugarcane cultivars response to orange rust in greenhouse
Sugarcane orange rust, caused by Puccinia kuehnii (Krüguer) Butler, is considered one of the most important diseases, due to losses in agricultural yields in susceptible commercial plantations. The objective of this wo...
Maturation and germination of somatic embryos of Sorghum bicolor (L.) Moench cultivar ‘CIAP 132R-05’
In sorghum [Sorghum bicolor (L.) Moench], developed protocols for plant regeneration via somatic embryogenesis do not include maturation stage. The present work was carried out with the aim of achieving the maturation a...
