Molecular methods for identification of monophasic Salmonella Typhimurium strains

Abstract

Two molecular biology methods were used to differentiate [i]Salmonella enterica[/i] 1,4,[5],12:i:- strains: “[i]Salmonella[/i] Check&Trace microarray” (CT) and multiplex PCR (mPCR). For 92 strains in CT result “[i]Salmonella[/i] 1,4,[5],12:i:-” were obtained. Those strains were confirmed in mPCR as monophasic[i] fljB[/i]-lack [i]Salmonella[/i] Typhimurium. For 17 strains, which in CT assay were recognized as [i]Salmonella[/i] Typhimurium, the same identification was obtained in mPCR. Reference [i]Salmonella[/i] strains: Lagos, Agama, Tsevie, Glocester and Tumodi in CT were recognized as [i]Salmonella [/i]genovar, in mPCR – as [i]Salmonella[/i] O:4, H:i other than [i]Salmonella[/i] Typhimurium, the same like [i]Salmonella[/i] Farsta, recognized incorrectly in CT as [i]Salmonella [/i]Typhimurium.
