Protocol for the isolation of intact chloroplasts from leaves Musa spp. obtained by in vitro culture and evaluation of photosynthetic activity

Abstract

Chloroplasts are organelles of plant cells where photosynthesis takes place by converting light energy into chemical energy (ATP) and generating reducting power (NADPH). Isolation of chloroplasts and evaluation of photosynthetic activity in vitro has been a difficult task due to structural and functional changes suffered by chloroplasts during their isolation and the presence of contaminating cell fractions. In this paper we propose a protocol for the isolation of chloroplasts from banana leaves and a procedure for the evaluation of the photosynthetic activity of chloroplast suspension. The main reagents, solutions, biological materials, equipment, and conditions necessary to reproduce it in laboratory conditions are summarized. Finally, several applications of this protocol are suggested to be used in teaching and scientific research related to plant biotechnology.
