Separation/purification of DNA by solid phase extraction with 330 anion exchange resin

Abstract
A solid-phase extraction procedure for the separation/purification of DNA from complex matrix was developed with anion exchange resin 330 as sorbent. DNA was adsorbed onto the resin by electrostatic interaction which was afterwards collected by stripping with Na2HPO4 as eluent. Some of the experimental parameters governing the sorption and desorption of DNA have been investigated, including sample pH, adsorption time and temperature, stripping reagent and concentration. A sorption capacity of 5.98 g/mg for DNA was achieved under the optimal conditions. The DNA separated/purified from a synthesized sample and the human whole bloods exploiting the developed procedure were used as templates for PCR successfully.
