SOLUBILITY PROFILES OF SELECTED DRUGS IN EDIBLE OIL MEDIA

Abstract
The solubility of selected drugs in edible oils was determined to promote development of edible oil based dosage forms and to initiate an oil solubility data base. Aqueous based formulations are open to hydrolytic, oxidative, catalytic and microbial degradation resulting in short shelf lives. Oil based Ayurvedic preparations have much longer shelf lives. Aspirin, diazepam, paracetamol and phenytoin were subjected to solubility studies in pure sesame oil, pure olive oil and in two series of co-solvents of each of these two oils with oleic acid, tocopherol and castor oil. All of these were blended by a ratio of 5 parts in 100 parts of the solvent. Twelve edible oil vehicles were studied for each drug. Maximum solubility was, for aspirin 3.76 mg/ml in a co-solvent of sesame oil:oleic acid:tocopherol:castor oil 85:5:5:5 parts, diazepam 10 mg/ml and phenytoin 2.27 mg/ml each in pure sesame oil and paracetamol 0.42 mg/ml in a co-solvent of sesame oil:tocopherol 95:5 parts. Accordingly oil based effective dosage forms of diazepam are possible. Solubilities of phenytoin nearly 7 mg/3 ml and aspirin 12 mg/3 ml in respective vehicles could be pharmacologically effective in migraine and in cerebral blood clots when administered by the brain targeted transcranial route. The dissolution of non polar drugs in oil media is a tardy process taking several weeks. The comparatively limited solubilities detected indicate that the concept is more applicable to drugs effective in low doses
