STUDIES ON ALMOND GUM BASED COLON TARGETED TABLETS OF SECNIDAZOLE AND ITS β-CYCLODEXTRIN COMPLEX TO TREAT AMOEBIASIS

Abstract
The present investigation was planned to formulate colon targeted tablets of Secnidazole using almond gum as matrix carrier. Also, it was planned to improve the solubility of Secnidazole by forming inclusion complex with β-cyclodextrin. Different formulations were prepared by changing the concentrations of matrix carrier and β-cyclodextrin. Initially, granules were prepared and evaluated for various rheological properties like bulk density, tapped density, compressibility index and angle of repose and for compression characteristics like hardness, thickness and disintegration time by using standard techniques. In vitro release was conducted for all the formulations in USP XXIV for 24 h in different pH resembling different portions of gastro intestinal tract. Formulations containing 25% w/w almond gum showed maximum drug release in colonic environment. These formulations were complexed with β-cyclodextrin. After complexation of Secnidazole with β-cyclodextrin, Secnidazole dissolution is significantly increased in colonic environment. The colon targeted matrix tablet of Secnidazole showed no change and significant difference in physical appearance, drug content or in dissolution pattern after stability studies conducted at 400C / 75 % RH for 180 days.
