Tetramethylpyrazine improves postoperative neurocognitive disorders in old mice by attenuating blood-brain barrier injury

Abstract
Objective To investigate the effect and mechanism of tetramethylpyrazine ( TMP) on postoperative neurocognitive disorders after splenectomy in old mice. Methods Old mice were randomly divided into control + normal saline group (Sham group), model + normal saline group (Con group) and model + tetramethylpyrazine group (TMP group). TMP group was intraperitoneally injected with TMP (40 mg/ kg) for 7 d, and Sham group and Con group were intraperitoneally injected with equal dose of normal saline. The mouse model of neurocognitive disorders was established by splenectomy. The memory ability of mice was detected by novel object recognition test. The blood-brain barrier (BBB) permeability of mice was detected by Evans blue (EB) assay, the microscopic changes of BBB were observed by electron microscope, and the changes of neurons in hippocampal CA1 region were observed by hematoxylin-eosin ( HE) staining. Results Compared with Sham group, the discrimination index (DI) of mice in Con group was significantly decreased (P < 0. 05), EB content in brain was significantly increased (P < 0. 05), the BBB was significantly damaged under electron microscope, and HE staining showed obvious neuronal inflammation in hippocampal CA1 region. Compared with Con group, DI in TMP group was significantly increased (P < 0. 05), EB content in brain was significantly decreased (P < 0. 05), the BBB disruption was attenuated, and the neuronal inflammation in hippocampal CA1 region was alleviated. Conclusion TMP improves postoperative neurocognitive disorders in old mice after splenectomy by attenuating BBB disruption.
