The Antioxidant Activity of Palmitoleic Acid on the Oxidative Stress Parameters of Palmitic Acid in Adult Rat Cardiomyocytes

Abstract
Objectives Many researches have focused on reducing the deleterious effects of oxidative stress in biological systems. The present study investigated the effect of palmitoleic acid on the oxidative stress parameters induced by palmitic acid on cardiomyocytes. Methods Rat cardiomyocytes were treated with 0.5 mM palmitate, palmitoleate, and palmitate + palmitoleate, or remained as control. Total antioxidant capacity (TAC) and Malondialdehyde (MDA) values were measured in both the cells and the media at 24h and 48h intervals. Results While cellular TAC values decreased in the palmitic acid group compared to the control group (24%) at 48h time point (P < 0.001), the palmitoleic acid group showed 45% and 19% increases (P ≤ 0.001); Co-incubation of palmitoleate and palmitate indicated 44% (P = 0.001) and 11% (P = 0.02) increases compared to the control group at 24h and 48h time points, respectively. Co-incubation of palmitoleate and palmitate tended to decrease the MDA values in the cells and media compared to the palmitate group at 24h time point (P = 0.09). Conclusions In conclusion, the harmful effects of the oxidative stress induced by palmitate would be diminished by palmitoleate in the rat cardiomyocytes.
