The Restriction Site Amplified Polymorphism (RSAP) Used for Sex Identification of Osteoglossum Bicirrhosum

Abstract

The sex-specific molecular marker plays an important role in improving the process of artificial selective breeding in fish aquiculture. However, there are some fish which do not have obvious sexual dimorphism just like Osteoglossum bicirrhosum. In the present study, restriction site amplified polymorphism (RSAP) molecule marker technique was used for finding the sex-specific marker of O. bicirrhosum. Of the 45 primers screened, 43 produced highly reproducible fragments except for R1+R8 and R2+R5 primers. Although the RSAP markers were unable to distinguish sex of O. bicirrhosum in view of the limited samples, our study provided the first evidence that the RSAP technique was applicative for this species. Given its superiority, we suggest that successful application of RSAP to O. bicirrhosum will evidently enhance our capability in analyzing genetic diversity, linkage map and gene mapping involving O. bicirrhosum.
