COMPARATIVE STUDY ON VARIOUS TABLETING PROPERTIES OF PHENYTOIN SODIUM MOUTH DISSOLVING DOSAGE FORM WITH AND WIT HOUT SPHERICAL CRYSTALS

Abstract

The aim of the present research is to advance the engineering of pharmaceutical agglomerates, for the purpose of tailoring the properties by modulation of processing parameters like solvent composition, hydrodynamics, and supersaturation generation. Spherical Phenytoin Sodium crystals were produced by an agglomeration technique with the use of three solvents system namely Ethanol–Dichloromethane– Demineralized Water employing a three blade mechanical stirrer. Micromeritic properties of agglomerates such as bulk density, flow property, compatibility and cohesivity were optimized and their compression behavior was investigated and compared with that of raw crystals. Into effect of such modifications in the crystal habit, other parameters like, dissolution rate and stability were changed. Spherically agglomerated crystals were tableted as a mouth dissolving tablet by direct compression method without capping in a single-punch tableting machine and different parameters were evaluated. Decreased Carr’s index (13.3) and Hausner’s ratio (1.07) for spherical crystals indicated increased flow properties. The reduction in parameter ‘a’ in Kawakita equation and the increase in parameter ‘b’ compared with those of original drug crystals proved that the flowability and packability of agglomerated crystals were preferably improved for direct tableting.
