Design and characterization of nanoparticulate drug delivery system of an anticancer drug: Imatinib mesylate

Abstract
The present study was aimed to develop a nanoparticulate drug delivery system of Imatinib mesylate using a biodegradable polymer such as poly lactic acid (PLA). The design of Imatinib mesylate nanoparticles is proposed for better bioavailability by prolonged systemic circulation and effective drug delivery at the site of action at a controlled release. The work also experiments the possibility of using PEG as a copolymer to increase the specificity of the formulation by modifying the surface characteristics of nanoparticles. The Imatinib mesylate nanoparticles were prepared by Multiple emulsion solvent evaporation technique. The prepared nanoparticles were characterized with respect to Particle size, Zeta potential, sterility test, Drug entrapment efficiency, Invitro drug release profile, Pharmacological evaluation and stability studies. The formulation showed good entrapment and an extended drug release profile. The particle size and zeta potential values indicated that the particles remain separate and highly repelling. This repelling property was thought to be useful in decreasing Opsonization and favours target specificity. The pharmacological evaluation revealed that the Imatinib mesylate nanoparticle treated animals showed significant inhibition of tumor volume, packed cell volume and tumor (viable) cell count. The stability tests carried out for a period of 3 months at various storage conditions showed that the formulation remains stable throughout the period of study. The results indicate that the Imatinib mesylate loaded PLA/PEG blend nanoparticles posed to be a promising candidature for the effective drug delivery increasing target specificity of Imatinib mesylate in treatment of cancer.
