Effect of inoculum density and frequency of immersion on the in vitro propagation of Psidium guajava cv. Red dwarf in temporary immersion systems

Abstract
The objectives of the present work were to study the effect of the inoculation density and the immersion frequency in the multiplication of guava explants in temporary inmersion systems. Systems with flasks of 1.0 liter of capacity with 0.2 liters of culture medium were used. The best results were obtained when 20 explants for flask were inoculated and cultured with a frequency of four immersions a day and after 21 days the culture medium was renovated. After 42 days a multiplication rate of 44.7 was obtained. Hyperhidric explants were observed independently of the immersion frequency with a smaller affectation in the multiplicated explants in the treatment with four immersions per day. With the objective of achieving the elongation the explants were transferred to systems with flasks of 10 liters of capacity with 3.0 L of culture medium. They were cultured with four immersions per day during 21 days, lapsed this time the culture medium was renovated totally by 5.0 l of rooting culture medium. The number of immersions decreased to one per day and the effect of forced ventilation in the quality of the obtained explants was evaluated. During the rooting phase the multiplication rate increased (70.3), but under acclimatization conditions the explants survival was only achieved (21.8%) in the treatment with forced ventilation.
