Effect of light and gibberellic acid on in vitro germination of Capsicum annuum L. cv. ‘Papri King’

Abstract

In the coastal zone of Peru, paprika (Capsicum annuum L.) is a crop of great importance, but it has a high production cost because the seeds are germinated in seedlings and this stage is considered crucial. The objective of the present investigation was to determine the effect of light and gibberellic acid on the in vitro germination of paprika seeds cv. ‘Papri King’. The seeds were disinfected with 70% alcohol for 1 min and then sodium hypochlorite (0.5, 1.0, 1.5 and 2.0%) during 10 minutes. For germination it were placed in MS culture medium. Treatments were formed with the addition of gibberellic acid (AG 3 ) (0.25, 0.5, 1.0 mg l -1 ) and photoperiod of 16 hours light or dark. The percentage of contamination and germination was evaluated in vitro in the first and fourth week and the height (cm) of the seedlings was measured. Using 1.5% and 2% sodium hypochlorite solutions, the seeds were disinfected without microbial contamination. The addition of gibberellic acid and the photoperiod of 16 h light increased the seeds germination. With 1 mg l -1 gibberellic acid and photoperiod of 16 hours light, the highest germination percentage was achieved (97.8%).
