Evaluation of tolerance to Finale ® in germination and regeneration in vitro of two Cuban varieties of rice (IACuba-17 and IACuba-19)
Journal Title: Biotecnología Vegetal - Year 2005, Vol 5, Issue 1
Abstract
Selection agent used during the shoot selection has an important role on the transgenic plant generation efficiency. In this work, the tolerance to the herbicide Finale ® in two Cuban rice cultivars, IACuba17 and IACuba-19 was evaluated, and determined that 10 days exposure to 5 and 10 mg.l -1 of Finale ® were enough to avoid seedlings of IAC-17 and IAC-19, respectively. Cultivated calluses (0, 2, 4 and 6 days) in the absence of Finale ® in the regeneration medium were used to evaluate the minimal concentration of Finale ® that totally inhibits shoot regeneration. Pre-induced calluses cultured during two days and 3 mg.l -1 of Finale ® in the regeneration medium was the most efficient combination to select shoots during the generation of transgenic plants resistant to the herbicide. Our shoot selection procedure reduces to 3 weeks the time to obtain shoots during the generation of transgenic rice plants.

Callogenesis in hybrid cultivars of Cocos nucifera L. by in vitro culture of immature inflorescences
In Colombia, coconut palm cultivation (Cocos nucifera L.) is centralized in the Pacific Coast and has productive limitations. An alternative to improve production is the use of hybrid cultivars. However, efficient meth...
Determination of the minimal inhibitory concentration of Geneticin G418 in the callus formation process of Digitalis purpurea L.
Digitalis purpurea L. produces cardiatonic glycosides such as digoxin and digitoxin, which are widely used for congestive heart failure. Genomic studies in this specie are limited because there is not an efficient genet...
Field response of Eucalyptus grandis W. Hill ex Maiden plants obtained by micropropagation and seeds
The use of plants obtained by in vitro culture could contribute to satisfy the demand of products derived from this plant in the agricultural sector. However, the response of plants in the field is not well known. The...
Somatic embryo differentiation and germination of Coffea arabica L. cv. Catimor 9722 obtained in an orbital shaker
The present work had as objectives to study the effect of four inoculation densities on the process of differentiation of embryogenic cell suspensions of Coffea arabica cv. Catimor 9722 and to evaluate the effect of two...
In vitro establishment of the hybrid rootstock ‘Garfi x Nemared’ (Garnem) for peach
The interspecific hybrid between almond and peach, ‘Garfield x Nemared’ (Prunus dulcis (Mill) D.A.Webb x Prunus persica (L.) Batsch.) has become very important as rootstocks for peach in Bolivia, but propagation by tra...
