Extraction of total RNA in plants and filamentous fungi

Abstract

RNA of high quality is a common element required as starting material in the majority of the techniques used in molecular studies either in plants or in phytopathogenic fungi. In this manuscript we emphasized in the characteristics of each of these tissues and the convenience to use certain protocols, which are going to be differentiated by the application of different detergents. We also dealt with possible solutions to eliminate the high contents of polysaccharides that difficult the total extraction of ARN. In addition, some RNA extraction protocols to obtain good yields during the interaction plant-filamentous fungi are mentioned. As common element, either for plants or filamentous fungi, a detailed description of the steps to be followed to obtain total ARN is done. Besides, the different variants when we face problems related to the tissues structure of this investigation were also studied.
