In vitro comparative evaluation of sealing ability of root canal sealers using glucose penetration test

Abstract

Introduction. Several studies on microleakage tests were published. The variety of evaluation methodologies and test parameters are major reasons for discrepancies, and it is hard to compare the results even when referring to one type of a sealer. In 2005, a new method for the analysis of endodontic microleakage was introduced, based on the filtration rate of glucose along the root canal filling. Aim of the study. To present in vitro evaluation of the sealing ability of three different sealers placed in root canals and the evaluation of a new glucose penetration method. Material and methods. Fifty single-rooted permanent human teeth, extracted for periodontal reasons, were selected for this study. The root canals were instrumented using the M-two system (VDW, Germany) with a single-length technique and filled with one of the tested materials: AH Plus Jet (Dentsply DeTrey, Germany), RSA RoekoSeal (Coltene Whaledent, Germany), Endomethasone N (Septodont, France) using a single-cone technique with greater taper. All the roots were then placed in a device for microleakage test. After 1, 14, and 28 days, the roots were analysed with a Glucose Assay Kit GAGO 20 (Sigma Aldrich, USA) and a spectrophotometer. Results. The greatest concentration of glucose was achieved after 1 day for AH Plus Jet and after 14 and 28 days for Endomethasone N. After 1 day of observation there were no statistically relevant differences in glucose concentrations among the study groups. After 14 and 28 days of observations, the concentration of glucose for AH Plus Jet and RSA RoekoSeal was lower than for Endomethasone N. Conclusion. The glucose concentration depended on the type of root canal sealer and the period of study. RSA RoekoSeal seemed to provide better sealing properties than other tested materials. The glucose penetration method was found to be effective in evaluation of root canal system microleakage.
