INHIBITORY EFFECTS OF SALINOMYCIN ON THE PROLIFERATION, CELL CYCLE AND MIGRATION OF HUMAN MELANOMA A375 CELLS IN VITRO

Abstract

Malignant melanoma (MM) is one of the deadliest cancers of any skin cancer, which develops through malignant transformation of melanocytes. It usually caused by DNA damage resulting from exposure to ultraviolet (UV) light from the sun, Genetics also play a role. It’s responsible of 80% of deaths from skin cancer, in the early stage, can be treat by resection of surgery, while melanoma in the late stage is difficult to treat with current therapies. In this thesis, we use Salinomycin to effect inhibits on the Melanoma malignant A375 cells. To investigate the effects of salinomycin in Melanoma malignant A375 cells, Salinomycin deficient cells were used in the study. The results showed that salinomycin deficiency inhibited cell proliferation and Migration of Melanoma A375 cells; however, use Immunofluorescence staining to investigate the effect of Salinomycin on the cytoskeleton F-actin of melanoma A375 cells. decreased cytoskeleton F-actin on Melanoma A375 cells. These results suggest that Salinomycin might contribute to the malignancy of tumor cells; therefore, Sal might be a potential prognosis factor for metastatic M A375 cancer.
