Naloxone precipitates nicotine abstinence syndrome and attenuates nicotine-induced antinociception in mice.
Journal Title: Pharmacological Reports - Year 2005, Vol 57, Issue 6
Abstract
The present study focused on the evaluation of a role of opioid system in nicotine-induced antinociception and physical dependence in mice. The results indicate that nicotine (3 mg/kg) produced a significant antinociception in the hot plate test. Additionally, the opioid receptor antagonist naloxone (0.5 and 1 mg/kg), dose-dependently attenuated this effect. Our second experimental protocol consisted in intermittent administration of nicotine (2.5 mg/kg, sc) four times daily for 7 days. In order to precipitate nicotine abstinence, mice were given one injection of mecamylamine (3 mg/kg) or naloxone (1 mg/kg) one hour after the last nicotine injection on the test day (day 8) in the morning. Interestingly, our findings revealed that both drugs precipitated somatic withdrawal signs in mice, with a slight difference in their influences on the intensity of several signs. These data support the hypothesis that similar opioid-cholinergic interactions are involved in nicotine-induced antinociception and nicotine withdrawal syndrome.

Reductive metabolism of the sanguinarine iminium bond by rat liver preparations.
Background: Sanguinarine (SA) is a quaternary benzo[c]phenanthridine alkaloid that is mainly present in the Papaveraceae family. SA has been extensively studied because of its antimicrobial, anti-inflammatory, antitumor,...
Antiproliferative activity of parthenolide against three human cancer cell lines and human umbilical vein endothelial cells.
Parthenolide is a major sesquiterpene lactone derived from feverfew (Tanacetum parthenium) with known anti-inflammatory activity. Moreover, the anticancer potential of this compound was suggested. In this study, we deter...
Bismuth increases hydroxyl radical-scavenging activity of histamine H(2)-receptor antagonists.
The effects of histamine H(2)-receptor antagonists, alone or in a combination with bismuth, on ( )OH-provoked degradation of deoxyribose were studied. The histamine H(2)-receptor antagonists (cimetidine, ranitidine and r...
Endothelial mitochondria as a possible target for potassium channel modulators
Variety of ion channels is present in plasma membrane of endothelial cells. These include the potassium channels such as Ca(2+)-activated K(+) channels, inwardly rectifying K(+) channels, voltage-dependent K(+) channels...
