Transcriptomic response of goat mammary epithelial cells to Mycoplasma agalactiae challenge – a preliminary study
Journal Title: Animal Science Papers and Reports - Year 2015, Vol 33, Issue 2
Abstract
Mycoplasma agalactiae (Ma) is one of the main aetiological agents of intramammary infections in small ruminants, causing contagious agalactia. To better understand the underlying disease patterns a primary goat mammary epithelial cell (pgMEC) culture was established from the mammary tissue and challenged with Ma. High-throughput mRNA sequencing was performed to reveal differentially expressed genes (DEG) at different time-points (3 h, 12 h, and 24 h) post infection (PI). The pathway enrichment analysis of the DEG showed that infection significantly affected pathways associated with immune response, steroid metabolism, fatty acid metabolism, apoptosis signalling, transcription regulation, and cell cycle regulation. Based on the results we suggest that mammary epithelial cells in vivo contribute to the immune system by the induced expression of cytokines and other chemotactic agents, activation of the complement system and apoptosis pathways, and expression of genes coding for antimicrobial molecules and peptides. In our study we attempted to interpret the detected transcriptomic changes in a biological context and infer mammary infection resistance candidate genes, interesting for further validation. Additionally, the results represent comprehensive goat mammary transcriptome information and demonstrate the applicability of the comparative genomics approach for annotation of goat data, using transcriptome information of a closely related species (Bos taurus) as a reference.

Power of different microsatellite panels for paternity analysis in sheep
The aim of this work was to develop and to test PCR-based suitable microsatellite marker panels for paternity testing in sheep. Blood samples were collected from 212 Kivircik sheep (24 rams and their 188 offspring) rais...
Polymorphism and structure of exon 2 of caprine melatonin receptor 1b gene and its relations to fertility and seasonal estrus
The first description is presented of nucleotide sequence of exon 2 of caprine melatonin receptor 1b (MT2). No polymorphisms of MT2 gene were detected between high fertility and year-round estrous goat breeds and low fer...
Emotional response of young race horses to a transfer from a familiar to an unfamiliar environment
The aim of this study was to measure emotional arousal in young race horses as expressed by heart rate (HR) during routine situations of grooming, hoof cleaning, and while at rest, having been transported from the enviro...
The effect of dietary oil seeds on the fatty acid profile and metabolism in ostrich liver
The study was carried out on 40 ostriches randomly allocated into five groups and each group was randomly assigned to a different dietary treatment. Experimental diets were made on the basis of a control diet, where a pa...
cDNA cloning of porcine CDH1 and its expression profile in porcine early parthenotes
The study aimed at cloning the complete cDNA sequence of porcine CDH1 gene and detecting its expression in different developmental stages of early parthenotes. After cloning the CDH1 gene from porcine oviduct using rapid...
