Sleep Spindles Induction by Cinnamaldehyde

Abstract
Background and purpose: Sleep spindles oscillations are usually defined as distinct waves having a frequency of 12- 20 Hz with almost 0.5-1 second duration and usually maximum amplitude over central brain regions. They are generated by interplay of the thalamic reticular nucleus (TRN) and other thalamic nuclei during stage 2 NREM sleep. It is believed that they have special role in memory consolidation. Cinnamaldehyde as an herbal medicine with positive effect on memory repair could be investigated in modulating sleep spindles. Materials and methods: In this study male Wistar rats (n=7) were subjected to orally treatment of Cinnamaldehyde (20mg/day/kg) and the other (n=7) consider as control group. The electroencephalogram (EEG) was recorded with stainless steel screw electrodes placed over the skull and the electromyogram (EMG) were recorded from dorsal neck muscles. The sleep stages were recorded during 2 hours for 3 days. Sleep spindles density were investigated by a semi-automatic sleep scoring method developed in MATLAB. Results: The finding showed Cinnamaldehyde (20mg/day/kg) treatment for 20 consecutive days could significantly (P <0.01) increase the sleep spindle density in comparison with control group which just received the vehicle. Conclusion: Cinnamaldehyde could improve sleep spindles and this may be a reason for its usefulness in memory repair.
